Structure
SS-31 (D-Arg-Dmt-Lys-Phe-NH₂) belongs to the Szeto-Schiller peptides. Alternating aromatic and basic residues and 2′,6′-dimethyltyrosine (Dmt) allow cell entry despite the positive charge.
Research focus
Studies examine stabilisation of cristae structure, the respiratory chain, formation of reactive oxygen species and models of mitochondrial, cardiac and renal disease. Elamipretide has been tested in several clinical trials.
Status
In 2025 elamipretide received accelerated approval in the USA for a rare mitochondrial disease; there is no authorisation in the EU.
